Comparative Distribution of Carbohydrates and Lipid Droplets in the Golgi Apparatus of Intestinal Absorptive Cells

چکیده
Complex carbohydrates have been demonstrated cytochemically in the Golgi apparatus of the rat intestinal absorptive cell by specific stains for acid mucopolysaccharides (4) and for glycoproteins (15) . A staining gradient in the cisternae was observed in these studies, increasing towards one face of the Golgi apparatus, generally referred to as the mature face . In addition, the early accumulation of labeled carbohydrates from radioactive precursors has suggested an involvement of the Golgi apparatus in the synthesis of this product for transport to other positions (2, 6, 9, 14) . In the absorptive cell, much of this carbohydrate is subsequently transported to and incorporated into the surface coat, especially that covering the microvilli . Lipid droplets are common in absorptive cells of animals on a normal diet. In the Golgi apparatus, these droplets are present in varying numbers, depending on the absorptive state of the cell, and they show a preferential accumulation in vacuolar dilations of the cisternae closest to the plasma membrane (7, 11) . There is some evidence to suggest that these droplets represent chylomicrons or lipoproteins formed during lipid absorption and destined for release from the cell along the lateral boundaries (5, 7, 10) . A simultaneous occurrence of carbohydrates and lipid droplets in the Golgi apparatus and their respective distributions have not been demonstrated previously. A bifunctional condition is presently described in which the lipid droplets and carbohydrates are processed primarily on opposing faces of the Golgi apparatus .

HDL-mediated cholesterol uptake and targeting to lipid droplets in adipocytes.
Adipocytes express high levels of the HDL scavenger receptor class B type I in a differentiation-dependent manner. We thus have analyzed the routes of HDL cholesterol trafficking at different phases of adipocyte differentiation in the 3T3-L1 cell line. One novel and salient feature of this paper is the observation of a widespread distribution in the cell cytoplasm of Golgi markers, caveolin-2, ...
متن کاملEffects of Puromycin on the Structure of Rat Intestinal Epithelial Cells during Fat Absorption
This report provides information on the morphology of rat intestinal epithelial cells during fat absorption. In addition, the role of protein metabolism in this process has been evaluated by blocking its synthesis with puromycin and studying the fine structure of mucosal cells from rats at various times after fat intubation. The results indicate that SER-derived vesicles, containing fat droplet...

The properties of the amoebocytes and intestinal epithelium of Venus mercenaria were studied by a variety of cytochemical procedures designed to demonstrate proteins, enzymes, carbohydrates, and lipids. The cytoplasm of the amoebocytes contains specific granules which are constantly present and which are interpreted as being atypical mitochondria. Identification of their mitochondrial nature re...

The microanatomy of the intestinal epithelium in the Chinese soft-shelled turtle (CST) was studied by light and transmission electron microscopy (TEM). The small intestinal epithelium (SIE) was single layered or pseudostratified. The enterocytes contained mitochondria or mitochondria and lipid droplets. The enterocytes were arranged tightly in the apical parts of epithelium and connected by des...
متن کاملSynthesis and transport of lipoprotein particles by intestinal absorptive cells in man.
The site of synthesis and some new details of lipoprotein particle transport have been demonstrated within the jejunal mucosa of man. In normal fasting volunteers, lipoprotein particles (88%, 150-650 A diameter) were visualized within the smooth endoplasmic reticulum and Golgi cisternae of absorptive cells covering the tips of jejunal villi. Electron microscopic observations suggested that thes...
